ABSTRACT
Enzim
is the protein that has function as catalyst for various chemistry reaction in
biology system. There are three enzims sources center that are, plants, animals
and microbas. The value of enzym that has been produced by microba always
increasing. It is caused microbas have some superiors such as, easily to grow
up and the production can be increased. Penicillin G Acylase (PGA) is a enzym
that can be catalysis hydrolysis penicillin G becomes 6 aminopenisilanat acid
(6-APA) which are representating the basic substance in making semisintetic
penicillin. In general bacterias produce that enzym as intracellular. According
to Sari’s research (2011) says that icolating bacterias from Buffalo’s
intestines (+) products PGA enzym. This research was done at Kopertis
Laboratorium Region X Padang on January 2013 and this is a continue research
that has purposed for determining pH optimum activities PGA that are produced
by icolating bacteria from Buffalo’s intestines. This research is experimental
research, that is by giving treatment for the variation of pH between 6,5-8,0
toward PGA activities that are produced by icolating the bacteria and calculate
of enzym activities which are produced by using Kornfeld method. Kornfeld
method has the high sencitivities and quickly in detecting acid 6-APA compound
that are the result of penicillin G hydrolysis by PGA enzym. The activities
(AE) of PGA enzym which are produced by isolating of bacterias from Buffalo’s
intestines show the high activities at pH 7,5 among AE 0,051 unit/ml enzym.
Declining activities of the enzym above or under pH optimum (7,5) caused enzym
conformation changing which are influential toward AE so that makes AE more low
than pH which under or up pH 7,5.
